Police say the driver is assisting them in their enquiries, and have asked anyone who witnessed the incident or may have information to get in touch.
The A38 Bristol Road remains closed in both directions from the Edithmead Roundabout, at junction 22 with the M5, and the Fox and Goose Pub located at the next junction north from the roundabout.
Those travelling north on the A38 after leaving the motorway are having to make diversions as police advise motorists to avoid the area.
Earlier this morning police confirmed the road remains closed due to a "serious collision" which is sadly now confirmed to be fatal.
A spokesperson for Avon and Somerset Police said: "Officers investigating a fatal collision on the A38 in Brent Knoll this morning (August 29) are appealing for witnesses to come forward.
"We were called by the ambulance service at 5.20am to the collision involving a pedestrian and a white Vauxhall Astra.
"Despite the best efforts of emergency services, the pedestrian, a man in his 40s, died at the scene. His family have been informed and our thoughts are with them."
The spokesperson added: "The driver of the car remained at the scene and has assisted officers with their enquiries.
"The A38 has been closed between the Fox and Goose pub and the B3140 roundabout while the scene investigation is completed."
Police are asking anyone who witnessed the collision or who has any information which could help their investigation to call 101 using the reference number 5226248501.
It is understood that the emergency services remain at the scene to ensure traffic safety while the closure is in place, and that members of the public have been advised by the police the road will remain closed this morning while enquiries are ongoing.
